Cryopreservation of bovine testicular tissue

Abstract

The cryopreservation of bovine testicular tissue is a viable alternative for the reproductive potential preservation. In addition to storing tissue fragments containing a large number of germ cells, it allows the preservation of the niche around these cells, offering physical support and mechanisms for the regulation of spermatogonial stem cells. This article aims to analyze the scientific productions related to the protocols for cryopreservation of bovine testicular tissue. It is an integrative literature review conducted in March 2023. The PICO strategy was used to formulate the guiding question: What is the scientific evidences available in the literature on the cryopreservation of bovine testicular tissue? The databases consulted were PubMed, Scopus, and Web of Science, using the descriptors cryopreservation, tissues, tissue, testis, testicle, and cattle. Articles available in full and without language and publication date restrictions were selected. In total, twelve articles were analyzed, with a publication highlight between the years 2015 and 2016, most of them were developed in China and published in the journal Cryobiology. It was observed that all research used the technique of uncontrolled slow freezing. The Eagle's Modified Medium supplemented with cryoprotectants, such as dimethyl sulfoxide, glycerol, and ethylene glycol, was more commonly used for cryopreservation. Regarding the size of the fragments, they varied from 1 - 2 mm to 0.3 - 0.5cm3. The standardization of cryopreservation protocols for bovine testicular tissue remains a challenge to be overcome due to the difficulty of harmonizing the different protocols, in addition to factors such as exposure time and cryoprotectant solution.
