Viability of using papain in the treatment of wounds contaminated by Staphylococcus aureus in Wistar rats

Abstract

Papain, a protease of plant origin, has been used for decades as an alternative for the treatment of wounds associated with infectious processes. This study aimed to evaluate healing by second intention of skin wounds contaminated and treated with 10% papain. Fifteen healthy male rats were used, which were submitted to two incisions: Control Wound treated with 0.9% saline solution and Test Wound, treated with 10% papain ointment. Both were previously contaminated by inoculation of Staphylococcus aureus. The animals were divided into three groups with five rats each: G1 - biopsied on the 3rd day, G2 - 7th day, and G3 - 14th day after the wounds were performed. The evaluation of the wounds was based on macro and microscopic aspects in the predetermined periods. After the biopsy, histological sections stained by Hematoxylin Eosin and Masson's Trichrome were performed. It was macroscopically verified that 10% papain in the Test Wound was more efficient when compared to Control at the 7th and 14th days of the experiment. Microscopically, Test Wounds had a better response, compared to Control Wounds, in several aspects: less presence of hematofibrinous crust; less inflammatory infiltrate on the 3rd day and 7th day, greater amount of granulation tissue ranging from moderate to severe on the 14th day; greater amount of fibroblasts on the 7th and 14th day, and greater presence of collagen matrix (organized) on the 14th day. Thus, it was concluded that the use of 10% papain in contaminated tissue lesions can positively assist the healing process.
